Analytical Method Development and Validation for Simultaneous Estimation of Metoprolol Tartrate and Hydrochlorothiazide

Abstract:

Purpose: To develop a simple, easy and isocratic reversed phase high-performance liquid chromatography technique for concurrent estimation of metoprolol tartrate and hydrochlorothiazide. Methodology: The desired chromatographic separation was achieved by using Agilent C18 column (ODS UG 5 column, 250 mm×4.5 mm). The mobile phase consisted of a mixture of phosphate buffer (pH 3.0): acetonitrile: methanol in the ratio of 60:30:10 (v/v) at a flow rate of 1.0 mL/min and detected at 237 nm. Findings: For both the compounds, the calibration curves showed a good linear correlation over the concentration range of 10-50 and 5-25 μg/mL of metoprolol tartrate and hydrochlorothiazide respectively. The retention times were 3.37 min and 6.40 min for metoprolol tartrate and hydrochlorothiazide respectively. The LOD was found to be 0.33 μg/ml and 0.16 μg/ml and LOQ was found to be 1.01 and 0.04 μg/ml for metoprolol tartrate and hydrochlorothiazide respectively. Conclusion: A simple, easy and isocratic reversed phase high-performance liquid chromatography technique has been developed for concurrent estimation of metoprolol tartrate and hydrochlorothiazide. The developed method has been validated according to ICH guidelines.
